摘 要 目的:利用随机引物聚合酶链反应探讨血链球菌各菌种基因分型,比较它们之间的基因差异。方法:设计随机引物5′AAG AGA GGAGCT AGC TCT TCT TGG 3′,提取血链球菌标准菌株5株和临床菌株8株的染色体DNA后进行PCR扩增。结果:血链球菌各菌种扩增的基因片段有较大区别,且实验稳定性、重复性较好,可用于细菌的鉴定。结论:随机引物聚合酶链反应可用于血链球菌各菌种的鉴定和分型。

Abstract:Objective:To study gene identification of Streptococcus sanguis group (SSG) strains and compare their differences by arbitrarily primed polymerase chain reaction (APPCR).Methods:APPCR DNA polymorphism technique was used to distinguish SSG strains by designing arbitrary primer 5′ AGA
GGA GCT AGC TCT TCT TGG 3′ and extracting chromosomal DNA of 13 SSG strains including model and clinical species.Results:There were great differences in the main band of polymorphism among SSG species.The experiment was steady and can be repeated.Conclusion:APPCR DNA polymorphism method can rapidly identify and classify SSG species.

血链球菌属口腔原生菌群,与维持口腔生态平衡密切相关。它们可通过唾液获得性薄膜粘附于牙齿表面,为其它细菌提供结合位点,导致一些病理过程的发生,如龋病。一些口腔卫生和治疗措施还可引起菌血症,血链球菌就通过血液循环引起全身性疾病如感染性心内膜炎。有些学者还认为血链球菌对牙周可疑致病菌有拮抗作用,是牙周的有益菌群。并建议运用血链球菌进行牙周病的细菌治疗。根据目前分类[1],原称血链球菌其实是由一组细菌组成:可分为口腔链球菌,缓症链球菌、高氏链球菌、血链
